Generation of H7N9-specific human polyclonal antibodies from a transchromosomic goat (caprine) system.

To address the unmet need for good human polyclonal antibodies as therapeutic and diagnostic reagents, building on the previous platform was established transchromosomic (Tc) of our cattle, we report here the development of goat Tc system expressing Cavia Clia Kits human polyclonal antibodies in their sera. In Tc goats system, man-made chromosome (HAC), which consists of the entire gene repertoire of human immunoglobulin (Ig) in the germline configuration are introduced into the genetic makeup of the domestic goat. We achieved this by transferring the HAC to the goat fetal fibroblast cells followed by somatic cell nuclear transfer for the production of goat Tc.

Analysis of gene and protein expression in cells of peripheral blood mononuclear (PBMC) and sera, respectively, Tc caprine successful expression of human Ig genes and antibodies. In addition, immunization with inactive Tc caprine influenza A (H7N9) virus H7N9 Hemagglutinin followed by 1 (HA1) improving human antibodies induce high neutralizing activity against H7N9 virus in vitro. As a small hoofed animals, Tc caprine offers the advantages of low cost and rapid establishment of livestock, therefore complement Tc cattle platform in response to various needs of medical and diagnostic applications where a small volume of products necessary human antibodies.

This communication addresses the research hypothesis that part of the iso 17: 0 and anteiso 17: 0 fat milk may come from the synthesis of endogenous extraruminal network. In order to confirm this linear regression model was applied to calculate the proportion of iso 17: 0 and anteiso 17: 0 in the milk fat that can come from lengthening their alleged precursors iso 15: 0 and anteiso 15: 0, respectively. Sixteen dairy goats were allocated to two simultaneous experiments, in a crossover design with four animals per treatment and two 25 d experimental periods.

In both trials, alfalfa hay is the only feed and forage to concentrate ratio (33: 67) remains constant. different experimental diet on the composition of the concentrate, both rich in starch or neutral detergent fiber, and they are given alone or in combination with 30 g / d of linseed oil. Iso 15: 0, anteiso 15: 0, iso 17: 0 and anteiso 17: 0, the branched-chain fatty acid that is most abundant in milk fat, determined by gas chromatography using Chicken Clia Kits two different capillary columns. The regression model determined that 49% of iso 17: 0 and 60% of anteiso 17: 0 fat milk extraruminally formed from iso 15: 0 and anteiso 15: 0 elongation.

Although infant formula is important breastmilk substitutes, have been shown to contain many different lipid structures compared to human milk. In this study, lipidome and fatty acid composition of the human, and caprine dairy cows were analyzed and compared. A total of 13 classes of lipids (including TG, DG, SM, PC, Cer, HexCer, Hex2Cer, PE, PG, PS, PI, PA and CL) were analyzed.

Breast milk is richer in TG containing LA, SM contains ULCFA and PL containing ARA, DHA, and DGLA. Moreover, there is no differential between the DG and BC human species and caprine milk. A total of 215 and 147 lipids identified as a potential biomarker that could be used to further analyze the differences in the biological nature of man, cattle and dairy caprine
